以频哪醇和化合物(CAS: 1238702-56-1)为原料合成1-苯基-吡唑-5-硼酸频那醇酯的一般步骤如下:参考例132,将1-苯基-5-(4,4,5,5-四甲基-1,3,2-二氧杂硼杂环戊烷-2-基)-1H-吡唑与1-苯基-1H-吡唑-5-硼酸的混合物在室温下溶于甲苯(86 mL)中。随后,加入频哪醇(5.39 g,45.6 mmol)和硼酸(8.57 g,45.6 mmol)。将反应混合物加热至40℃,并在此温度下搅拌反应2天。反应完成后,将混合物在减压下浓缩,残余物用己烷研磨,得到1-苯基-吡唑-5-硼酸频那醇酯(7.93 g,收率64%),为浅黄色固体。产物经1H NMR(DMSO-d6, 300 MHz)表征:δ 1.23(12H,s),6.84(1H,s),7.34-7.59(5H,m),7.75(1H,d,J = 1.9 Hz)。
