以5-氯吡啶-2-羧酸(1.0g,6.35mmol)为原料,将其与浓HCl混合。向该混合物中加入H2SO4(0.1ml)和EtOH(10ml),随后在80℃下加热反应16小时。反应完成后,冷却反应混合物,并进行减压浓缩。将浓缩后的残余物溶解于EtOAc(50ml)中,依次用饱和NaHCO3溶液(25ml)和盐水(25ml)洗涤。有机相经MgSO4干燥后,过滤并减压浓缩,得到目标化合物5-氯吡啶甲酸乙酯(990mg,收率84%)。LCMS分析显示:计算值MH+为186;实测值为98%(MH+)m/z 186,保留时间Rt=1.13min。1H NMR(250MHz,CDCl3)δppm:8.63-8.76(1H,m),8.10(1H,d,J=8.4Hz),7.82(1H,dd,J=8.5,2.4Hz),4.49(2H,q,J=7.2Hz),1.45(3H,t,J=7.2Hz)。
