以二碳酸二叔丁酯和D-焦谷氨酸乙酯为原料合成BOC-L-焦谷氨酸乙酯的一般步骤:将D-焦谷氨酸乙酯(5.1g,32.6mmol)溶于二氯甲烷(70ml)中,依次加入三乙胺(4.55ml,32.6mmol)、二碳酸二叔丁酯(14.2g,65.2mmol)和4-(二甲基氨基)吡啶(3.98g,32.6mmol)。将反应混合物在室温下搅拌1.5小时,通过薄层色谱(TLC)监测反应进程,确认反应完全。反应完成后,将反应液用水洗涤两次,再用盐水洗涤,有机相用无水硫酸钠干燥,过滤后减压浓缩。粗产物通过柱色谱法(洗脱剂比例为1:1的乙酸乙酯/石油醚)纯化,得到Boc-D-焦谷氨酸乙酯,为浅黄色粘稠油状物(7.98g,收率95%),静置后固化为固体。
