以N-CBZ-3-羟基吡咯烷为原料合成N-Cbz-3-吡咯烷酮的一般步骤:将中间体6.iii(1.10 g)溶解于二氯甲烷(DCM,8 mL)中,冷却至0℃。在搅拌下,缓慢滴加N,N-二异丙基乙胺(DIPEA,2.5 mL),随后加入三氧化硫吡啶络合物(1.79 g)的二甲基亚砜(DMSO,6.5 mL)溶液。反应混合物在0℃下持续搅拌1小时,之后通过加入水(6 mL)淬灭反应。水相用乙醚/正己烷(1:1,3 × 5 mL)混合溶剂萃取,合并有机相。有机相经真空浓缩后,得到的残余物用乙醚/正己烷(1:1)混合溶剂溶解,并通过硅胶柱色谱法(正己烷/乙酸乙酯,5:5)纯化,得到1.05 g(96%收率)淡黄色油状产物N-Cbz-3-吡咯烷酮。产物经1H NMR(DMSO-d6;δ ppm)表征:2.48-2.61(2H,m);3.61-3.80(4H,m);5.09(2H,s);7.27-7.41(5H,m)。
