4-氯-3-硝基苯甲酸甲酯(1)的合成步骤:将4-氯-3-硝基苯甲酸(5.0g,24.8mmol)溶解于200mL甲醇中,加入4.15mL(29.8mmol)三乙胺。在冰盐浴中冷却反应混合物,缓慢滴加3.19mL(44.7mmol)乙酰氯。反应混合物在回流条件下搅拌6小时。反应完成后,通过减压蒸馏去除溶剂。残余物用100mL水溶解,然后用乙酸乙酯(100mL)萃取两次。合并有机相,用无水硫酸镁干燥,减压浓缩。粗产物用乙醚(10mL)重结晶,得到白色晶体4-氯-3-硝基苯甲酸甲酯(4.81g,收率92%)。薄层色谱(TLC)Rf = 0.55(展开剂:乙酸乙酯/环己烷 = 7/3);熔点79-80℃。红外光谱(IR)显示羰基吸收峰位于1716 cm-1;1H核磁共振谱(300MHz,DMSO-d6)δ3.90(单峰,3H,OCH3),7.90(双峰,1H,H5,J5-6 = 8.1Hz),8.15(双双重峰,1H,H6,J6-5 = 8.1Hz,J5-2 = 1.5Hz),8.49(双峰,1H,H2,J2-6 = 1.5Hz)。分子式:C8H6ClNO4。
