4-(4-氟苯甲酰基)丁酸的合成步骤如下:在2L三颈圆底烧瓶中加入250g无水AlCl3(1.87摩尔)和300mL氟苯(307.5g; 3.2摩尔),将混合物在冰浴中冷却至5℃。通过加料漏斗在45分钟内缓慢加入100g戊二酸酐(0.86mol)溶于400mL氟苯(4.3摩尔)的浑浊悬浮液,保持反应温度低于12℃。反应混合物逐渐升温至室温并搅拌90分钟,通过NMR监测反应完成。将反应混合物冷却至0至5℃,小心加入700mL 1N HCl的冷水溶液以淬灭未反应的AlCl3,初期保持温度低于20℃,后期不超过40℃。将混合物倒入2L 1:1的水和冰混合物(v/w)中沉淀粗产物,过滤白色悬浮液并用水充分洗涤。将白色固体溶于3L 5% NaHCO3饱和水溶液中,蒸汽浴加热1小时后趁热通过硅藻土垫过滤。滤液冷却至室温,逐滴加入约320mL浓HCl调节至pH 1,产物结晶析出,冰浴中搅拌30分钟。过滤,冰冷水洗涤湿滤饼,50℃真空干燥16小时,得到143.2g 4-(4-氟苯甲酰基)丁酸,熔点141至142℃,产率79.3%。
