以5-溴-2-氯苯甲醚为原料合成5-溴-2-氯苯酚的一般步骤:在0-5℃的搅拌条件下,将BBr3(8.2 mL,84.9 mmol)缓慢滴加到5-溴-2-氯苯甲醚(18.26 g,82.4 mmol)的二氯甲烷(45 mL)溶液中。反应混合物在室温下持续搅拌4小时。反应完成后,将混合物小心倒入预先冷却的2N NaOH/冰混合物中,并用叔丁基甲基醚(TBME)洗涤两次。水相用2N HCl酸化至pH值适宜后,再次用TBME萃取两次。合并所有有机相,用无水Na2SO4干燥,随后在减压下蒸发溶剂,得到目标产物5-溴-2-氯苯酚,为无色晶体(16.35 g,收率95%)。产物经1H-NMR(400 MHz,DMSO-d6)和质谱(ES+)分析确认:1H-NMR δ(ppm):6.97(dd,1H),7.09(d,1H),7.26(d,1H),10.65(bs,1H,OH);MS(m/z)ES+:210(15),208(70,M+),206(50),179(20),177(15),63(100)。
