6-羟基烟酸乙酯的制备:向6-羟基烟酸(5.0 g,35.9 mmol)的无水乙醇(180 mL)溶液中缓慢加入浓硫酸(1.0 mL)。将反应混合物于60℃下搅拌20分钟,随后加入额外的乙醇(33.0 mL),并将反应体系加热至回流状态,持续搅拌6.5小时。反应完成后,将反应液冷却至0℃,并缓慢加入饱和碳酸氢钠水溶液以中和反应液。随后,将乙醇通过旋转蒸发仪减压浓缩除去。用乙酸乙酯萃取水相,合并有机相,用饱和食盐水洗涤,无水硫酸钠干燥。过滤后,将滤液减压浓缩,得到白色晶体状产物6-羟基烟酸乙酯(5.33 g,收率89%)。产物经1H-NMR(CDCl3)表征:δ 1.36(3H,t,J = 7.1 Hz),4.33(2H,q,J = 7.1 Hz),6.58(1H,d,J = 9.5 Hz),8.01(1H,dd,J = 2.4, 9.5 Hz),8.19(1H,d,J = 2.4 Hz),12.43(1H,brs)。
