以甲醇和6-羟基烟酸为原料合成6-羟基烟酸甲酯的一般步骤:首先,将6-羟基烟酸(100g,719mmol)悬浮于甲醇(1L)中。随后,加入18M硫酸(50mL),并将反应混合物加热回流16小时。反应完成后,冷却反应混合物,缓慢加入碳酸氢钠粉末(45g)以部分中和酸性。接着,在减压条件下蒸除大部分甲醇。向残余物中加入水(1L),并小心加入碳酸氢钠溶液调节pH至7。用二氯甲烷(4×200mL)萃取悬浮液,合并有机相,用无水硫酸钠干燥。减压除去溶剂后,将所得固体在50℃的真空烘箱中干燥1.5小时,得到83g(产率75%)6-羟基烟酸甲酯,为白色固体。质谱(电喷雾离子化):154.2(MH+,C7H7NO3)。1H NMR(300MHz,CDCl3):δ 3.88(s,3H),6.59(dd,1H),8.02(dd,1H),8.21(m,1H),13.19(bs,1H)。
