以5-溴-3,4-二氢异喹啉-1(2H)-酮(4.3 g,18.9 mmol)和2,3-二氰基-5,6-二氯-1,4-苯醌(8.6 g,37.9 mmol)为原料,在1,4-二恶烷(76 mL)中混合。将反应混合物在100℃下搅拌24小时。反应完成后,蒸发去除溶剂,将残余物溶解于乙酸乙酯(500 mL)中,并用10%氢氧化钠水溶液(2×500 mL)洗涤。分离有机层和水层,水层进一步用乙酸乙酯(4×300 mL)萃取。合并所有有机层,用无水硫酸钠干燥,然后蒸发浓缩。产物通过快速色谱法纯化,洗脱剂为二氯甲烷与甲醇的混合溶剂(比例从99:1渐变至96:4),得到5-溴异喹啉-1(2H)-酮(1.49 g,6.65 mmol,收率35%),为黄色固体。产物经LCMS分析显示纯度为94%,保留时间(Rt)为1.243分钟,电喷雾质谱(ESMS)显示分子离子峰m/z为224([M+H]+)。
