首先,以购自TCI(日本)的反式-4-(氨基甲基)环己烷羧酸为起始原料,合成反式-4-(氨基甲基)环己烷乙基羧酸盐酸盐。在0℃条件下,将反式-4-(氨基甲基)环己烷羧酸(1.57g,10mmol)溶解于40mL乙醇中,并持续搅拌。缓慢滴加亚硫酰氯(0.9mL,12mmol)至反应体系中,保持低温并继续搅拌10分钟。随后,将反应混合物升温至70℃,回流反应1小时。反应完成后,将混合物冷却至室温。在减压条件下彻底蒸除溶剂,所得残余物用己烷(25mL)洗涤两次以去除残留溶剂。最后,将产物在真空条件下干燥6小时,得到白色固体反式-4-(氨基甲基)环己烷乙基羧酸盐酸盐,产量为2.13g(产率96%)。产物经1H NMR(d6-DMSO)表征:δ= 4.03(q,J = 7.08,2H,OCH2CH3),2.61(m,1H,H13),2.21(m,1H,H10),1.85(m,2H,H9),1.27(m,4H,H12, H11),1.16(t,J = 7.06,3H,OCH2CH3),0.96(m,4H,H12, H11)。元素分析结果(C10H20ClNO2)计算值:C,54.17; H,9.09; N,6.32。实测值:C,53.78; H,8.91; N,6.34。
