以11H-茚并[1,2-b]喹喔啉-11-酮(4a,0.23g,1.0mmol)为原料,与盐酸羟胺(0.35g,5.0mmol)在乙醇(EtOH,5.0mL)中混合,回流反应6小时(反应进程通过薄层色谱法(TLC)监测)。反应完成后,将混合物冷却至室温,并倒入水中。通过过滤收集生成的沉淀物,并用乙醇重结晶,得到目标产物11H-茚并[1,2-b]喹喔啉-11-酮肟(5a,0.23g,收率94%),为白色固体。熔点:274-276℃(在乙醇中重结晶,分解)。核磁共振氢谱(1H NMR,400MHz,DMSO-d6):δ 7.67-7.72(m,2H,ArH),7.78-7.85(m,2H,ArH),8.10-8.13(m,2H,ArH),8.14-8.17(m,1H,ArH),8.50-8.53(m,1H,ArH),13.48(s,1H,NOH)。核磁共振碳谱(13C NMR,100MHz,DMSO-d6):δ 122.26, 128.76, 129.30, 129.87, 129.99, 130.58, 132.09, 132.56, 133.07, 135.99, 141.56, 141.85, 147.18, 150.81, 152.89。电喷雾质谱(ESIMS):[M+H]+ m/z 248.01。元素分析计算值(C15H9N3O·0.2H2O):C 71.81%,H 3.78%,N 16.75%;实测值:C 71.47%,H 3.65%,N 16.80%。

体外研究

Compound IQ-1S is a potent, noncytotoxic inhibitor of pro-inflammatory cytokine [interleukin (IL)-1α, IL-1β, IL-6, IL-10, tumor necrosis factor (TNF)-α, interferon-γ, and granulocyte-macrophage colony-stimulating factor] and nitric oxide production by human and murine monocyte/macrophages. The effect of IQ-1S is evaluated on LPS-induced cytokine production in human PBMCs. Among the 12 cytokines analyzed, LPS (200 ng/mL) consistently induces five (IL-1α, IL-1β, IL-6, IL-10, and TNF-α) in PBMCs compared with DMSO-treated control cells. Production of all of these cytokines is significantly inhibited by 20 μM IQ-1S. Among them, TNF-α production is inhibited completely by IQ-1 (>99%), the levels of IL-1α, IL-1β, and IL-10 are decreased by 85%, and IL-6 production is decreased by 33%.

体内研究

When mice are dosed with 12.5 and 30 mg/kg IQ-1S (The sodium salt of IQ-1S) i.p., the serum exposure of the compound is also good, with AUC 0-12h values of 2.9 and 7.4 μM/h, respectively.
