在氮气氛下,将4-羟基苯甲腈(III)(5.0g,0.042mol)溶解于乙腈(50mL)中。将反应混合物冷却至-15℃,缓慢加入三氟甲磺酸(CF3SO3H)(3.7mL,6.3g,0.042mol)。维持反应温度在-15℃,分批加入N-溴代琥珀酰亚胺(NBS)(8.2g,0.046mol),共分6次加入。加毕,将反应混合物恢复至室温,继续在氮气氛下搅拌4小时,反应进程通过薄层色谱(TLC)监控(展开剂:己烷/乙酸乙酯=75:25,检测方法:UV和KMnO4显色)。当原料完全消耗后,用碳酸钠(Na2CO3)水溶液稀释反应混合物,并用甲基叔丁基醚(MTBE)(3×50mL)萃取。合并有机层,用无水硫酸钠干燥,减压浓缩除去溶剂,得到3-溴-4-羟基苯甲腈(IV),为白色固体(7.7g,收率93%)。1H NMR(DMSO-d6,300MHz,300K):δ=8.04(d,J=2.2Hz,1H),7.63(dd,J=2.2Hz,8.5Hz,1H),7.04(d,J=8.5Hz,1H)。
