在氮气保护和室温条件下,将3,4-二甲基吡啶(5.0g,46.6mmol)溶于浓硫酸(50.0mL)中。随后,分批加入N-溴代琥珀酰亚胺(9.31g,51.32mmol)。将反应混合物加热至60℃并搅拌18小时。反应完成后,将混合物冷却至室温,缓慢倒入冰冷的水中,并用10%氢氧化钠溶液调节pH至碱性。用乙酸乙酯(3×50mL)萃取水相。合并有机相,用饱和食盐水洗涤,无水硫酸钠干燥,减压浓缩得到粗产物。粗产物通过硅胶柱色谱(100-200目)纯化,以5%乙酸乙酯的己烷溶液为洗脱剂,得到3,4-二甲基-5-溴吡啶(0.8g,收率9.3%)。质谱(ESI)m/z:186.0 [M(79Br)+H]+,188.0 [M(81Br)+H]+;核磁共振氢谱(1H NMR,CDCl3):δ 2.31(s,3H),2.36(s,3H),8.23(s,1H),8.51(s,1H)。
