以4-氯-2-三氟甲基嘧啶-5-羧酸乙酯(55.7g)为起始原料,加入10%钯碳催化剂(0.3g)、乙醇(1000ml)及N,N-二异丙基乙胺(90ml)的混合物。在1大气压的氢气氛围下振荡反应2小时。反应完成后,过滤去除催化剂,并蒸发除去溶剂。将残余物溶解于二氯甲烷中,依次用氯化铵溶液和水洗涤,经无水硫酸镁干燥后,蒸发溶剂,得到目标产物2-(三氟甲基)嘧啶-5-甲酸乙酯,为浅黄色固体(48g,收率100%)。产物结构经1H-NMR(CDCl3)δ9.42(2H,s),4.51(2H,q),1.45(3H,t)及13C-NMR(CDCl3)δ162.7,159.4(2C),159.3(q,J=37Hz),126.3,119.6(q,J=275Hz),62.9,14.4确认。
