向(S)-1-BOC-吡咯烷-3-甲酸(863mg,4.01mmol)和K2CO3(829mg,6.00mmol)的无水DMF(8mL)溶液中加入碘甲烷(300μL,4.82mmol)。将反应混合物在室温下搅拌4小时。反应完成后,加入水,并用乙醚(Et2O)萃取三次。合并有机相,用无水硫酸镁(MgSO4)干燥,过滤后减压浓缩。残余物通过快速柱色谱法纯化(洗脱剂:正己烷/乙酸乙酯 = 3:1),得到目标产物(S)-1-Boc-3-羧基吡咯烷甲酯(916mg,100%),为无色油状物。产物经1H NMR(400MHz,CDCl3)确认:δ3.70(3H,s),3.65-3.28(4H,m),3.10-2.98(1H,m),2.18-2.05(2H,m),1.45(9H,s)。LRMS(ESI,[M + Na]+)显示分子离子峰为252.1。
