步骤A:甲基嘧啶-2-羧酸酯的制备:在0℃下,将干燥的氯化氢气体通入甲醇(700 mL)中30分钟,制备饱和氯化氢甲醇溶液。向此溶液中加入嘧啶-2-甲腈(21.585 g,205.38 mmol),室温搅拌反应16小时,随后升温至40-50℃继续搅拌3小时。反应完成后,减压浓缩反应混合物,将残余物溶解于水中。用固体碳酸氢钠调节水相pH至7.0。用20%异丙醇/二氯甲烷混合溶剂(3次)萃取水相。合并有机相,用无水硫酸钠干燥,过滤后减压浓缩,得到白色固体产物甲基嘧啶-2-羧酸酯(23.0 g,收率81%)。产物经1H NMR(400 MHz, CDCl3)表征:δ 8.97-8.96(d, J = 4.7 Hz, 2H),7.53-7.50(t, J = 4.7 Hz, 1H),4.09(s, 3H)。
