以2,5-二溴-3-甲基吡啶(M-12)(5.02g,20.0mmol)为原料,将其溶解于乙腈(50mL)中。随后,向该溶液中依次加入碘化钠(12.0g,80.0mmol)和乙酰氯(2.85mL,40.0mmol)。将反应混合物在回流条件下加热搅拌24小时。反应完成后,冷却至室温,加入适量水稀释,并用饱和碳酸氢钠水溶液调节pH至8以中和反应混合物。用乙酸乙酯进行萃取,收集有机层并用无水硫酸钠干燥,过滤除去干燥剂。减压浓缩除去溶剂,所得残余物通过硅胶柱色谱法(洗脱剂比例:正己烷/乙酸乙酯=98:2→70:30)进行纯化,得到目标产物5-溴-2-碘-3-甲基吡啶(M-13)(5.76g,产率97%),为红色油状物。
