以对溴溴苄和亚磷酸三乙酯为原料合成4-溴苄基膦酸二乙酯(16)的一般步骤:在圆底烧瓶中加入1-溴-4-(溴甲基)苯(5.0 g,20 mmol)和亚磷酸三乙酯(51 mL,300 mmol),混合物在90℃下回流反应19小时。反应完成后,通过减压蒸馏除去过量的亚磷酸三乙酯。粗产物通过快速柱色谱法(洗脱剂比例为1:1的己烷/乙酸乙酯)进行纯化,得到目标化合物16,产率为98%。产物为无色液体;1H NMR(400 MHz,CDCl3)δ 7.30(d,2H,J = 7.5 Hz),7.05(d,2H,J = 7.6 Hz),3.99-3.88(m,4H),2.99(s,1H),2.94(s,1H),1.12(t,6H,J = 6.9 Hz);13C NMR(100 MHz,CDCl3)δ 131.7, 131.6, 131.5, 121.0, 62.3, 34.0, 32.0, 16.5;HRMS计算值(C11H16BrO3P,[M + H]+)307.0097,实测值307.0093。
