以3-羟基吡啶为起始原料合成2-碘-3-羟基吡啶的一般步骤:向3-羟基吡啶(5.0g,52.57mmol)的水(400mL)溶液中依次加入碳酸钠(Na2CO3,11.6g,109.72mmol)和碘(I2,13.3g,52.57mmol)。将反应混合物在室温下搅拌2小时。反应完成后,用1M盐酸(HCl)溶液调节pH至4。过滤收集沉淀,用冷水洗涤,干燥后得到2-碘-3-羟基吡啶(10.7g,产率92%),为白色粉末。产物表征数据如下:熔点189-190℃(neat);薄层色谱(TLC)Rf = 0.33(展开剂为二氯甲烷/乙醇,97/3,v/v);红外光谱(IR,KBr压片)νmax(cm-1):2851, 2722, 2573, 2345, 1560, 1455, 1301, 1189, 1121, 794, 668;核磁共振氢谱(1H NMR,400MHz,DMSO-d6):δ10.88(s,1H,OH),7.87(dd,1H,J1 = 1.6Hz,J2 = 4.4Hz,H-6),7.24(dd,1H,J1 = 4.4Hz,J2 = 8.0Hz,H-5),7.18(dd,1H,J1 = 1.6Hz,J2 = 8.0Hz,H-4);核磁共振碳谱(13C NMR,100MHz,DMSO-d6):δ154.0, 141.8, 124.3, 121.3, 110.9;高分辨质谱(HRMS)计算值C5H4INO [M + H]+ 221.9410,实测值221.9416。
