向搅拌的DL-脯氨酸（1g，8.69mmol）的甲醇（10mL）溶液中缓慢加入亚硫酰氯（0.75mL，10.43mmol）。将反应混合物加热至回流温度并持续搅拌16小时。反应进程通过薄层色谱（TLC）进行监测。反应完成后，将反应混合物浓缩，残余物用二氯甲烷（15mL）溶解。加入固体碳酸氢钠以淬灭过量的亚硫酰氯，过滤后，将滤液减压浓缩，得到(R)-吡咯烷-2-羧酸甲酯（1.1g，收率98%）为淡黄色固体。
