以2-(1-苄基哌啶-4-亚基)乙酸乙酯为原料合成4-哌啶乙酸乙酯的一般步骤:向2-(1-苄基哌啶-4-亚基)乙酸乙酯(24g,93mmol)的乙酸乙酯(300mL)溶液中加入钯/碳催化剂(3.94g)。将反应混合物置于H-立方氢化反应器中,设定反应条件为55℃和55psi,进行氢化反应16小时。反应完成后,过滤除去催化剂,减压浓缩滤液,得到4-哌啶乙酸乙酯,为无色油状物(15g,收率90%)。产物经1H NMR(400MHz,CDCl3)表征,化学位移δ:4.10(m,2H),3.03(d,J = 16.4Hz,2H),2.59(t,J = 16.4Hz,2H),2.19(d,J = 9.6Hz,2H),1.87(m,1H),1.67(d,J = 17.2Hz,2H),1.23(t,J = 9.6Hz,3H),1.10(m,2H)。
