通用方法:在室温下,将LiOH·H2O(1.5当量)分批加入到酯类化合物2a-2b(1.0当量)溶解于THF(10体积)和H2O(10体积)的搅拌溶液中,持续搅拌反应混合物2小时。反应进程通过TLC监测。反应完成后,减压浓缩去除THF,用KHSO4水溶液酸化至酸性,随后用5%甲醇和二氯甲烷混合溶剂萃取。合并有机相,用无水Na2SO4干燥,过滤后减压浓缩,得到目标固体产物3a-3b。 4.2.3 2-(1,3-二甲基-2,6-二氧代-2,3-二氢-1H-嘌呤-7(6H)-基)乙酸(3a)的合成:将化合物2a(5g,18.78mmol)溶于100mL THF:H2O(1:1)混合溶剂中,加入LiOH·H2O(1.18g,28.18mmol),按上述通用方法操作,得到白色固体产物3a(4.1g,收率91%)。1H NMR(400MHz,DMSO-d6)δ:13.27(brs,1H),8.04(s,1H),5.07(s,2H),3.44(s,3H),3.20(s,3H);m/z(ES)+:239.04 [M+H]+。
