以2-溴-4-氟苯甲酸和碘甲烷为原料合成2-溴-4-氟苯甲酸甲酯的一般步骤:在23℃下,将DBU(1.7当量,1.16 mL,7.76 mmol)加入到含有2-溴-4-氟苯甲酸(1.0当量,1.0 g,4.57 mmol)和碘甲烷(2.0当量,0.57 mL,9.14 mmol)的乙腈(11 mL)溶液中。反应混合物在室温下搅拌48小时后,用去离子水(10 mL)淬灭反应,并用乙醚(2×10 mL)萃取。合并有机相,依次用去离子水(20 mL)和饱和食盐水(20 mL)洗涤,无水硫酸钠干燥后过滤。减压浓缩溶剂,所得粗产物通过快速柱层析(硅胶,95/5 正己烷/乙酸乙酯)纯化,得到无色油状标题化合物2-溴-4-氟苯甲酸甲酯1.0 g,收率98%。薄层色谱Rf = 0.50(展开剂:95/5 正己烷/乙酸乙酯);1H NMR(400 MHz, CDCl3, 25℃)δ: 7.88(dd, J = 8.8, 6.0 Hz, 1H), 7.41(dd, J = 8.3, 2.5 Hz, 1H), 7.08(ddd, J = 8.8, 7.6, 2.5 Hz, 1H), 3.92(s, 3H)ppm。
