步骤1(化合物34-2的合成) (0217)将L-苹果酸(80 g,600 mmol)溶解于无水乙醇(500 mL)中,在0℃下缓慢滴加亚硫酰氯(100 mL,1.5 mol)。将反应混合物在0℃下搅拌20分钟,随后升温至室温并继续搅拌过夜。反应完成后,通过减压浓缩去除溶剂,残余物用乙酸乙酯(1000 mL×3)萃取。合并有机相,依次用水(500 mL×2)和饱和氯化钠溶液(500 mL×2)洗涤,有机层用无水硫酸钠干燥,过滤后浓缩。粗产物通过硅胶柱色谱法纯化(洗脱液系统:石油醚/乙酸乙酯 = 20:1),得到(S)-2-羟基琥珀酸二乙酯(113 g,产率99%)。 (0218)1H NMR(400 MHz,CDCl3)δ:4.47(q,J = 4.8 Hz,1H),4.26(dq,J = 2.4, 7.2 Hz,2H),4.16(q,J = 7.2 Hz,2H),3.28(d,J = 4.8 Hz,1H),2.72-2.89(m,2H),1.20-1.34(m,6H)。
