以二碳酸二叔丁酯和反式-4-羟基-L-脯氨酸甲酯盐酸盐为原料合成N-Boc-反式-4-羟基-L-脯氨酸甲酯的一般步骤如下:将步骤I中得到的反式-4-羟基-L-脯氨酸甲酯盐酸盐(14.5g,0.1mol)悬浮于CH2Cl2(400mL)中,冷却至0℃,搅拌下依次加入Et3N(28mL,0.2mol)、DMAP(0.61g,5mmol)和Boc酐(27.5mL,0.12mol)。反应混合物逐渐升温至室温并继续搅拌2小时。反应完成后,减压浓缩除去溶剂,向残留固体中加入乙醚,过滤收集固体并用乙醚充分洗涤。合并滤液,减压浓缩。将残余物溶解于CH2Cl2中,依次用饱和NaHCO3水溶液和饱和NaCl水溶液洗涤,有机相用无水Na2SO4干燥。减压蒸除溶剂,得到浅黄色油状物,经高真空处理固化。所得固体用己烷多次研磨,高真空干燥,得到1-叔丁基-2-甲基(2S,4R)-4-羟基吡咯烷-1,2-二甲酸酯,为白色固体(24.5g,收率100%)。产物表征数据如下:(M+1) 245; 1H NMR (CDCl3, 300MHz) δ 4.54-4.37 (m, 2H), 3.75 (s, 3H), 3.70-3.40 (m, 2H), 2.48-2.21 (m, 2H), 2.13-2.0 (m, 1H), 1.46 (s, 3H), 1.41 (s, 6H)。
