N-[6-氯-5-(2-甲氧基苯氧基)[2,2'-二嘧啶]-4-基]-4-叔丁基苯磺酰胺性质、用途与生产工艺
生产方法
在装有机械搅拌器、温度计、回流冷凝器、滴液漏斗和加热套的烧瓶中,加入4,6-二氯-5-(2-甲氧基苯氧基)-2,2'-联嘧啶(105 g)和4-(叔丁基)苯磺酰胺(64.1 g,0.30 mol)的DMSO(525 mL)溶液。向该悬浮液中加入碳酸钾(83 g,0.60 mol)。在搅拌下将反应混合物加热至100-105℃,维持3-4小时。反应进程通过HPLC监测。反应完成后,将溶液冷却至20-25℃。在10-15分钟内缓慢滴加水(875 mL),继续搅拌1小时。过滤收集沉淀的固体,用水(2×350 mL)洗涤,随后转移至研钵中。加入水(800 mL)和浓盐酸(50 mL),搅拌30分钟。再次过滤收集沉淀的固体,依次用水(2×200 mL)和丙酮(2×175 mL)洗涤。将固体在40±2℃下减压干燥16-24小时,得到乳白色固体产物N-[6-氯-5-(2-甲氧基苯氧基)[2,2'-二嘧啶]-4-基]-4-叔丁基苯磺酰胺,产量为157.8-158.7 g(产率100%)。
