Uses
9-Methoxycanthin-6-one is a canthin-6-one alkaloid from the intact plant parts and callus cultures of Eurycoma longifolia (Tongkat Ali). Canthin-6-one alkaloids were evaluated for their cytotoxic activity against a HT-1080 human fibrosarcoma cell line.
Definition
ChEBI: An indole alkaloid that is the 9-methoxy derivative of canthin-6-one. Isolated from Eurycoma longifolia and Simaba multiflora, it exhibits cytotoxic activity towards human cancer cell lines.
Synthesis
1kg of bitter wood herbs were crushed and extracted with 10 liters of 75% ethanol by heating for 2 times, each time for 2 hours, combined with the extract, vacuum recovery of ethanol at 60 , the pH was adjusted to 11 with concentrated ammonia, extracted with equal volume of ethyl acetate for 2 times, combined with the extract, recovered the ethyl acetate, the residue was dissolved with 0.5% hydrochloric acid, and then the above steps were repeated with the concentration of ammonia to adjust pH - - ethyl acetate extraction. The total alkaloid was 17.1g. 600g of column chromatography silica gel (200-300 mesh) was loaded with chloroform, 10g of total alkaloid was dissolved in anhydrous ethanol, and 15g of column chromatography silica gel (100-120 mesh) was mixed with samples, and the samples were eluted with chloroform-methanol gradient after sample elution (starting solvent was chloroform), and the samples were collected in 250ml. Once, TLC check, combined with the same items, recovery of solvent, the total alkaloids of refined bitter wood 4.1g, high performance liquid chromatography (HPLC) method, 4,5-dimethoxy ferrocyanidinone, 4-methoxy-5-hydroxy ferrocyanidinone, the total content of the two according to the weight/weight of 83% (w/w). 600g of column chromatography silica gel (200-300 mesh) with chloroform wet method to load the column, take 10g of total alkaloids dissolved in anhydrous ethanol, and then dissolve the total alkaloids into the water. Alkaloids dissolved in anhydrous ethanol, 15g column chromatography silica gel (100-120 mesh) mixing samples, on the sample to chloroform methanol gradient elution (starting solvent for chloroform), 250ml collected once, TLC check, combined with the same item, repeated silica gel column chromatography, can be obtained 4,5-dimethoxy ferrocephalos 2.1g, HPLC normalization check purity of 98.7%; can be obtained 4-methoxy-5 4-methoxy-5-hydroxyferricyclohexanone 1.7g, HPLC normalization check purity 98.2%.
