Description

Midodrine is a prodrug of the α1-adrenergic receptor agonist, 1-(2’,5’-dimethoxyphenyl)-2-aminoethanol (Kis= 2, 6.9, and 1.7 μM for α1A, α1B, α1D, respectively). After oral administration it is completely absorbed via the intestinal H+-coupled peptide transporter 1 and converted to the active form through cleavage of a glycine residue. At 1-5 mg/kg, midodrine can produce a dose-related increase in mean arterial pressure in normotensive rats and improve the orthostatic index in an experimental model of postural hypotension.
