| Application | 3-(benzyloxymethyl)cyclobutanone can be used as a pharmaceutical intermediate in the synthesis of other compounds with certain activities. Examples of its applications are as follows: It can be used to prepare cis-3-[(benzyloxymethyl)cyclobutanol]: At -70°C, with stirring in tetrahydrofuran, a 1M solution of trisec-butylborohydride in tetrahydrofuran (40 ml) is added dropwise to a solution of 3-(benzyloxymethyl)cyclobutanone (1.166 g, 6.13 mmol), maintaining the reaction temperature below -65°C. The reaction is then heated to room temperature over 18 hours. The reaction mixture is quenched with a saturated aqueous sodium bicarbonate solution (25 ml) and then cooled to 5°C. A 30% aqueous hydrogen peroxide solution (4 ml) is added dropwise, maintaining the reaction temperature below 10°C. The mixture is extracted from water into ethyl acetate (50 ml), and the combined organic phases are washed with brine (30 ml), dried with magnesium sulfate, filtered, and evaporated under vacuum. The crude substance was purified by silica gel column chromatography and eluted with 25-50% ethyl acetate:pentane to give a colorless oil. |
