BUSERELIN Usage And Synthesis
Preparationcombined solid-phase and liquid-phase peptide synthesis method was employed. Using acid-labile 2-chlorotriphenylchloro resin (2-CTC resin), N,N-diisopropylcarbodiimide (DIC) and 1-hydroxybenzotriazole (HOBT) were used as condensing agents to gradually condense and obtain the busereline intermediate peptide resin. The intermediate was then cleaved with a low-concentration acid to obtain a fully protected busereline intermediate. In the liquid phase, ethylamine was added to achieve N-terminal modification. Catalytic hydrogenation was then used to remove the nitro group from the arginine side chain and the benzyl group from the tyrosine side chain. A suitable acid concentration was selected to remove the triphenylmethyl protecting group while retaining the tert-butyl group of D-serine to obtain the crude peptide. Finally, semi-preparative purification and lyophilization yielded high-purity busereline.
DescriptionBuserelin (Suprefact) is a hormonal therapy drug used to treat advanced prostate cancer. It is a medication which is used primarily in the treatment of prostate cancer and endometriosis. It is also used for other indications such as the treatment of premenopausal breast cancer, uterine fibroids, and early puberty, in assisted reproduction for female infertility, and as a part of transgender hormone therapy. In addition, buserelin is used in veterinary medicine. The medication is typically used as a nasal spray three times per day, but is also available for use as a solution or implant for injection into fat.

BackgroundBuserelin belongs to the group of gonadotrophin releasing hormone (gonadorelin) analogues (LHRH agonist). It acts on the pituitary gland which controls the amount of many different types of hormones (chemical messengers). It alters the amount of hormones, particularly the oestrogens androgens. This alteration of hormone levels can be exploited to treat cancers of the prostate gland, which are stimulated to grow by testosterone. Buserelin lowers the levels of testosterone, which starves the tumour of testosterone and causes it to shrink.
