Sodium Camptothecin (5 μM; 10 min) causes a severe inhibition of heterogeneously sedimenting nuclear RNA (HnRNA) synthesis.
Sodium Camptothecin also blocks a specific step in the processing of ribosomal precursor RNA, allowing the conversion of 45S RNA to 32S RNA, but inhibiting the conversion of 32S RNA to 28S RNA .

在无菌圆底烧瓶（250 mL）中准确称取1.045 g (S)-4-乙基-4-羟基-1H-吡喃并[3',4':6,7]吲哚嗪并[1,2-b]喹啉-3,14(4H,12H)-二酮（CPT内酯）。加入相当于20%最终体积的水，使CPT的最终浓度为25 mM。随后，向不均匀的混合物中加入3.34 g 1N NaOH溶液，确保CPT与NaOH的摩尔比为1:1.05。将混合物在50°C下剧烈搅拌至完全溶解，此过程约需1小时，表明CPT内酯已定量转化为其水溶性羧酸盐形式。研究表明，将温度提高至90°C可进一步加速这一转化过程。虽然搅拌4-6小时对CPT的化学稳定性无显著影响，但为确保反应完全，建议维持此搅拌时间。反应完成后，将溶液冷却至室温，并可在光照条件下于4°C保存至少24小时。（关于CPT-Na储备溶液的长期储存稳定性将在后续讨论。）最终溶液的pH值应保持在10至11.5之间。
