General procedure for the synthesis of 2-chloro-6-methoxypyridin-4-amine from methanol and 4-amino-2,6-dichloropyridine: 2,6-dichloropyridin-4-amine (41.6.A, purchased from Aldrich, 3.10 g, 19.0 mmol) was dissolved in 20% NaOMe/MeOH solution (15 mL), and the reaction was carried out at reflux for 72 hours. After completion of the reaction, the reaction mixture was diluted with H2O (40 mL) and extracted with 30% iPrOH/CHCl3 (3 × 30 mL). The organic layers were combined and washed sequentially with water (2 × 20 mL) and brine (15 mL), then dried with MgSO4. After removal of the organic solvent under reduced pressure, the residue was purified by rapid chromatography on silica gel with 0-10% MeOH/CH2Cl2 as eluent to afford the target product 2-chloro-6-methoxypyridin-4-amine (41.6.B) as a white solid (2.45 g, 81.2% yield).
