向500 mL水中加入氢氧化钠（97%，15.7 g，381 mmol），室温下搅拌悬浮液10分钟。加入6-甲基-2-硫杂亚砜-1,2,3,4-四氢嘧啶-4-酮（98%，53.5 g，369 mmol），搅拌混合物直至完全溶解，持续10分钟。缓慢滴加碘甲烷（28.99 mL，461 mmol），室温下继续搅拌反应混合物4小时。反应完成后，过滤得到无色固体，用冰冷的水（2×100 mL）洗涤，60℃下真空干燥，得到6-甲基-4-羟基-2-甲硫基嘧啶（57 g，收率98%），为无色固体。产物经LC-MS分析：m/z = 156.9 [M + H]+；保留时间（RT）= 0.61分钟。
