| Synthesis | N-{5-[2-(methylthio)ethoxy]pyrimidin-2-yl}hexanamide (6.8 g, 24 mmol) was used as a raw material and suspended in methanol (68 mL). To this suspension was added a methanolic solution of sodium methanolate (1 M, 120 mL, 120 mmol) and heated in an oil bath at 60 °C until complete dissolution. Subsequently, the reaction solution was stirred at the same temperature for 2 hours. After completion of the reaction, the reaction mixture was concentrated under reduced pressure. The residue was extracted with chloroform and water, and the organic layer was separated and washed with saturated brine. The organic layer was dried over anhydrous sodium sulfate and concentrated under reduced pressure. The resulting residue was washed with a mixed ether-hexane solvent and filtered to give 5-[2-(methylthio)ethoxy]pyrimidin-2-amine (2.93 g, 59% yield) as a light yellow solid. | | References | [1] Patent: US2009/62306, 2009, A1.
