Jingzhaotoxin III is a selective blocker (IC50=348 nM) peptide of the NaV1.5 channel. It is a cyclic peptide composed of 36 amino acids and contains three pairs of disulfide bonds, which have no effect on other subtypes, including NaV1.2, NaV1.4, NaV1.6, and NaV1.7. It is believed that the activation of sodium channels can be inhibited by binding to the NaV1.5 S3-S4 junction of domain II. Jingzhaotoxin III can selectively inhibit the activation of cardiac sodium channels, but has no effect on the sodium channels of dorsal root ganglion neurons.
