The CALB2 antibody targets calretinin, a calcium-binding protein encoded by the CALB2 gene, which belongs to the EF-hand calcium-binding protein family. Calretinin (CR) contains six EF-hand domains and regulates intracellular calcium signaling by buffering free calcium ions and modulating calcium-dependent processes. It is widely expressed in specific neuronal populations of the central and peripheral nervous systems, including the retina, cerebral cortex, hippocampus, and sensory ganglia. Beyond neurons, CALB2 is also found in non-neural tissues, such as mesothelial cells and certain tumors.

CALB2 antibodies are critical tools in immunohistochemistry (IHC) to identify calretinin-expressing cells, aiding in neuroanatomical studies and pathological diagnoses. In neuroscience, they help map neuronal subtypes and investigate neurodegenerative diseases linked to calcium dysregulation. In oncology, CALB2 antibodies assist in distinguishing mesothelioma (calretinin-positive) from adenocarcinoma (typically negative), supporting differential diagnosis. These antibodies are typically monoclonal or polyclonal, validated for techniques like Western blotting, IHC, and immunofluorescence. Specificity is crucial, as calretinin shares homology with other calcium-binding proteins (e.g., calbindin-D28k), necessitating rigorous validation to minimize cross-reactivity. Overall, CALB2 antibodies serve as versatile biomarkers in both research and clinical diagnostics.
