BUB1 (Budding Uninhibited by Benzimidazoles 1) is a serine/threonine kinase crucial for the spindle assembly checkpoint (SAC), a surveillance mechanism ensuring accurate chromosome segregation during mitosis. It localizes to kinetochores and plays dual roles: recruiting other SAC components (e.g., BUBR1. CDC20) to delay anaphase until chromosomes achieve proper microtubule attachment, and promoting chromosome alignment through Aurora B kinase regulation. Dysregulation of BUB1 is linked to chromosomal instability, aneuploidy, and cancer progression.

BUB1 antibodies are essential tools for studying mitotic checkpoint mechanisms, protein localization, and expression levels in cell cycle research. They are widely used in techniques like Western blotting, immunofluorescence, and immunohistochemistry to detect BUB1 in human, mouse, or other model organisms. Commercial antibodies often target conserved regions, such as the N-terminal or kinase domains. Validation includes testing in knockout cell lines or siRNA-treated samples to confirm specificity.
