Anillin (ANLN) is a conserved actin-binding protein crucial for cytokinesis and cell cycle regulation. First identified in *Drosophila*, ANLN plays a pivotal role in organizing actin filaments and septins during contractile ring assembly, ensuring proper cleavage furrow formation and abscission in dividing cells. Structurally, it contains an N-terminal PH domain for membrane binding and a C-terminal actin-binding region. ANLN expression peaks during the G2/M phase and is tightly regulated; its dysregulation is linked to various cancers, including breast, lung, and pancreatic cancer, where overexpression correlates with poor prognosis, metastasis, and therapeutic resistance.

ANLN antibodies are essential tools for studying cytokinesis mechanisms, cell cycle dynamics, and cancer biology. They are widely used in techniques like Western blotting, immunofluorescence, and immunohistochemistry to detect ANLN localization, expression levels, and interactions with cytoskeletal components. In research, these antibodies help elucidate ANLN's dual role as both a scaffold protein and a regulator of RhoA signaling. Clinically, ANLN is explored as a potential biomarker for cancer diagnosis or prognosis. However, antibody specificity remains a challenge due to ANLN's structural homology with other actin-binding proteins, necessitating careful validation in experimental settings.
