CLSPN (Claspin) is a critical protein involved in the DNA damage response and cell cycle checkpoint regulation. It acts as a scaffolding molecule that facilitates the activation of the ATR-Chk1 signaling pathway, which is essential for maintaining genomic stability during DNA replication stress. Claspin interacts with phosphorylated ATR (ataxia-telangiectasia and Rad3-related) kinase and recruits it to sites of stalled replication forks, enabling the phosphorylation and activation of checkpoint kinase 1 (Chk1). This cascade halts cell cycle progression, allowing time for DNA repair before entering mitosis.
