Red fluorescent protein (RFP) antibodies are immunological tools designed to detect and localize RFP-tagged proteins in experimental systems. RFP, originally derived from the coral *Discosoma* sp., is a β-barrel-structured fluorescent protein that emits red light (λ~558-583 nm) when excited. Its development expanded the color palette of fluorescent markers beyond green fluorescent protein (GFP), enabling multicolor imaging and improved tissue penetration in microscopy.

RFP antibodies are typically generated by immunizing hosts (e.g., rabbits, mice) with recombinant RFP or its variants (e.g., mCherry, tdTomato). These antibodies exhibit high specificity for RFP epitopes, allowing applications in Western blotting, immunofluorescence, flow cytometry, and immunohistochemistry. They are particularly valuable in tracking RFP-fusion proteins to study subcellular localization, protein-protein interactions, or gene expression dynamics in live or fixed cells.

The utility of RFP antibodies extends to validating CRISPR/Cas9-edited cell lines (confirming RFP knock-in/knockout) and analyzing transgenic organisms engineered with RFP reporters. Commercial RFP antibodies are often validated for cross-reactivity with common RFP derivatives, though batch-specific verification remains critical. As red fluorescence minimizes autofluorescence interference in biological samples, RFP-based systems paired with specific antibodies continue to advance research in cell biology, neuroscience, and developmental studies, offering spatial and temporal resolution in complex experimental models.
