The BAG4 antibody targets the BAG family molecular chaperone regulator 4 (BAG4), also known as SODD (Silencer of Death Domains), a protein involved in regulating apoptosis and stress responses. BAG4 belongs to the BAG protein family, characterized by their BAG domain, which interacts with heat shock protein 70 (HSP70) to modulate protein folding, degradation, and anti-apoptotic signaling. BAG4 specifically binds to death domains of tumor necrosis factor (TNF) receptor 1 (TNFR1) and related receptors, inhibiting apoptosis by blocking death domain-mediated signaling. It also interacts with Bcl-2. further promoting cell survival.

BAG4 is implicated in cancer progression, where its overexpression is linked to tumor growth, chemotherapy resistance, and poor prognosis in cancers such as breast and lung cancer. Additionally, BAG4 participates in NF-κB activation, inflammation, and endoplasmic reticulum (ER) stress responses. Antibodies against BAG4 are essential tools for studying its expression, localization, and functional roles via techniques like Western blotting, immunohistochemistry, and co-immunoprecipitation. Research on BAG4 antibodies may contribute to therapeutic strategies targeting apoptosis pathways or molecular chaperone networks in cancer and neurodegenerative diseases. However, its dual roles in pro-survival and stress adaptation require further exploration to clarify context-dependent mechanisms.
