AXIN1 (Axis Inhibition Protein 1) is a key scaffolding protein in the Wnt/β-catenin signaling pathway, which regulates embryonic development, cell proliferation, and tissue homeostasis. It acts as a tumor suppressor by facilitating the formation of the β-catenin destruction complex, comprising Adenomatous Polyposis Coli (APC), Glycogen Synthase Kinase 3β (GSK3β), and Casein Kinase 1 (CK1). This complex promotes β-catenin phosphorylation and subsequent proteasomal degradation, maintaining low cytoplasmic levels of β-catenin. Dysregulation of AXIN1. through mutations or reduced expression, disrupts this process, leading to β-catenin accumulation and aberrant activation of Wnt target genes, which is implicated in cancers like hepatocellular carcinoma and colorectal cancer.

AXIN1 antibodies are essential tools for studying its expression, localization, and interactions in both physiological and pathological contexts. These antibodies are widely used in techniques such as Western blotting, immunohistochemistry, and immunofluorescence to detect AXIN1 protein levels in tissues or cell lines. They also aid in elucidating post-translational modifications (e.g., phosphorylation) or mutations that affect AXIN1 stability or function. Additionally, AXIN1-targeting antibodies have potential therapeutic applications, particularly in cancers driven by Wnt pathway hyperactivation.
