HIP1R (Huntingtin-interacting protein 1-related protein) is a cytosolic protein involved in clathrin-mediated endocytosis and membrane trafficking. It belongs to the HIP1 protein family, sharing structural homology with HIP1. including an N-terminal ANTH (AP180 N-terminal homology) domain for lipid binding and a C-terminal talin-like domain for actin interaction. HIP1R binds to clathrin, actin filaments, and the huntingtin protein (HTT), playing a role in maintaining synaptic vesicle recycling and cytoskeletal organization. Dysregulation of HIP1R has been linked to neurodegenerative disorders, particularly Huntington's disease, due to its interaction with mutant HTT, which may impair endocytosis and contribute to neuronal toxicity.

HIP1R antibodies are essential tools for studying its expression, localization, and function in cellular processes. They are widely used in techniques like Western blotting, immunofluorescence, and immunohistochemistry to detect HIP1R in various tissues, including brain samples. These antibodies help elucidate HIP1R's role in disease mechanisms, such as abnormal protein aggregation in neurodegeneration or altered membrane trafficking in cancer. Validated HIP1R antibodies often target specific domains (e.g., ANTH or C-terminal regions) and are tested for cross-reactivity in human, mouse, and rat models.
