The DBF4 antibody is a research tool designed to detect and study the DBF4 protein, a key regulatory component in eukaryotic DNA replication. DBF4 (Dbf4-dependent kinase subunit) serves as the regulatory partner of the CDC7 kinase, forming the CDC7-DBF4 complex (also known as the DBF4-dependent kinase, DDK). This complex is essential for initiating DNA replication by phosphorylating subunits of the MCM (minichromosome maintenance) helicase, enabling the unwinding of DNA at replication origins during the G1/S phase transition of the cell cycle. DBF4 expression is tightly cell cycle-regulated, peaking during S phase and declining thereafter.

Antibodies targeting DBF4 are widely used in molecular and cell biology to investigate replication initiation mechanisms, cell cycle progression, and checkpoint responses. They enable techniques like Western blotting, immunofluorescence, and immunoprecipitation to analyze DBF4 protein levels, localization, and interactions. Dysregulation of DBF4 has been implicated in genomic instability and cancer, making these antibodies valuable in oncology research. For example, studies use DBF4 antibodies to explore overexpression in tumors or its role in replication stress responses.
