成纤维细胞活化蛋白（FAP/分离酶）是一种II型跨膜丝氨酸蛋白酶，其膜结合型与可溶型均具有脯氨酸后切割内肽酶活性，尤其偏好切割Ala/Ser-Gly-Pro-Ser/Asn/Ala共识序列；同时可降解明胶及热变性I型胶原，并展现二肽基肽酶活性（优先作用于Ala-Pro、Ile-Pro、Gly-Pro、Arg-Pro和Pro-Pro）。膜结合型FAP通过与DPP4、PLAUR或整合素结合，介导细胞外基质（ECM）的细胞周蛋白水解，进而促进细胞粘附、迁移及侵袭；在神经胶质瘤中通过降解蛋白聚糖短蛋白聚糖促进脑实质侵袭。值得注意的是，该蛋白在黑色素细胞中通过不依赖丝氨酸蛋白酶活性的方式调控增殖与存活，发挥抑癌作用。
