白细胞介素4(IL-4)是一种可诱导初始辅助T细胞(从Th0细胞向Th2细胞)分化的细胞因子。在Th2型免疫应答(尤其见于过敏反应与寄生虫感染过程中)产生的IL-4和IL-13共同作用下,巨噬细胞将发生差异性活化。该细胞因子是白细胞介素4受体的配体,而该受体亦能与IL-13结合,这可能是IL-4与IL-13功能存在诸多重叠的原因之一。STAT6(信号转导与转录激活因子)已被证实在介导该细胞因子的免疫调节信号中起核心作用。最新研究发现,在Itk、KLF2、CBP及Id3等多种基因缺陷型小鼠中,IL-4在胸腺内源性CD8+ T细胞的发育过程中起着关键作用,且该过程无需抗原预先刺激。
