髓系细胞表面抗原CD33亦称为SIGLEC3、Siglecs（唾液酸结合免疫球蛋白样凝集素）及GP67，是一种属于免疫球蛋白超家族和SIGLEC（唾液酸结合免疫球蛋白样凝集素）家族的单次跨膜I型膜蛋白。人源CD33/Siglec-3 cDNA编码的364个氨基酸多肽链包含一个疏水信号肽、一个N端免疫球蛋白样V型结构域、一个免疫球蛋白样C2型结构域、一个跨膜区及一个胞质尾区。CD33/Siglec-3通常被认为是髓系特异性标志物，但也在部分淋巴样细胞中表达。在免疫应答过程中，CD33/Siglec-3可通过其SH2结构域招募胞质磷酸酶，使信号分子去磷酸化从而阻断信号传导，经配体诱导的酪氨酸磷酸化后可作为抑制性受体发挥作用。CD33/Siglec-3能够诱导急性髓系白血病细胞发生凋亡。
