背景介绍
成纤维细胞活化蛋白（FAP，亦称为seprase）是一种II型跨膜丝氨酸蛋白酶。其细胞膜形式与可溶形式均具有脯氨酸后切割内肽酶活性，对Ala/Ser-Gly-Pro-Ser/Asn/Ala共识序列表现出显著偏好。能降解明胶及热变性I型胶原。同时具有二肽基肽酶活性，偏好底物为Ala-Pro、Ile-Pro、Gly-Pro、Arg-Pro及Pro-Pro。
细胞膜形式的FAP通过与DPP4、PLAUR或整合素结合，参与细胞外基质的细胞周围蛋白水解过程，从而促进细胞在细胞外基质中的粘附、迁移与侵袭。通过降解蛋白聚糖短蛋白聚糖，促进胶质瘤细胞在脑实质中的侵袭。在黑色素细胞中，FAP以不依赖于丝氨酸蛋白酶活性的方式，通过调控细胞增殖与存活发挥肿瘤抑制功能。
