研究背景：2-氧戊二酸脱氢酶复合物催化 2-氧戊二酸转化为琥珀酰辅酶 A 和 CO2 的全转化。该复合物含有三种酶成分的多个拷贝：2-氧戊二酸脱氢酶 (E1)、二氢硫辛酰胺琥珀酰转移酶 (E2) 和硫辛酰胺脱氢酶 (E3)。DLST（2-氧戊二酸脱氢酶复合物的二氢硫辛酰胺-残基琥珀酰转移酶成分，线粒体）也称为 DLTS 或 2-氧戊二酸脱氢酶复合物成分 E2，是一种 453 个氨基酸的蛋白质，属于 2-氧酸脱氢酶家族。DLST 与一种硫辛酰胺辅因子共价结合，并通过糖胺途径参与 L-赖氨酸降解。 DLST 定位于线粒体，形成具有八面体对称性的 24 个多肽结构核心。编码 DLST 的基因定位于人类染色体 14q24.3 和小鼠染色体 12 D2。

Proteins were transferred to a PVDF membrane and blocked with 5% nonfat powdered milk in PBST for 2 hour at room temperature. The primary antibody (1/2000) and Loading control antibody (GAPDH, 1/3000) was diluted with 5% nonfat powdered milk in PBST at 4°C overnight. Goat Anti-Rabbit IgG-HRP Secondary Antibody (1/8,000) was incubated for 45min at room temperature.
